Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000400.4(ERCC2):c.358G>C (p.Glu120Gln), citing Ambry Variant Classification Scheme 2023: The p.E120Q variant (also known as c.358G>C), located in coding exon 5 of the ERCC2 gene, results from a G to C substitution at nucleotide position 358. The glutamic acid at codon 120 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
