Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001242957.3(MAK):c.497G>A (p.Arg166His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 166 of the MAK protein (p.Arg166His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with retinitis pigmentosa (PMID: 21835304, 25385675). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 29784). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt MAK protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
