This variant lies in the TINF2 gene (transcript NM_001099274.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1062, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant has not been reported in the literature in individuals affected with TINF2-related conditions. This variant is present in population databases (rs767141183, gnomAD 0.006%). This sequence change affects an acceptor splice site in intron 6 of the TINF2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in TINF2 cause disease. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
