Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006445.4(PRPF8):c.3334C>T (p.Arg1112Cys), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with PRPF8-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.002%). This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 1112 of the PRPF8 protein (p.Arg1112Cys). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt PRPF8 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
