NM_002427.4(MMP13):c.1A>G (p.Met1Val) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects the initiator methionine of the MMP13 mRNA. The next in-frame methionine is located at codon 64. This variant is present in population databases (rs143446525, gnomAD 0.002%). Disruption of the initiator codon has been observed in individual(s) with clinical features of autosomal recessive metaphyseal dysplasia (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
