Pathogenic for Galactosylceramide beta-galactosidase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000153.4(GALC):c.166G>C (p.Asp56His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 56 of the GALC protein (p.Asp56His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with leukodystrophy (PMID: 31885218). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GALC protein function.
