Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000102.4(CYP17A1):c.1304T>C (p.Phe435Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 435 of the CYP17A1 protein (p.Phe435Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of CYP17A1-related conditions (PMID: 31695722; Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects CYP17A1 function (PMID: 31695722). For these reasons, this variant has been classified as Pathogenic.
