This variant lies in the TGFBR1 gene (transcript NM_004612.4) at coding-DNA position 1043, where G is replaced by A; at the protein level this means replaces cysteine at residue 348 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces cysteine, which is neutral and slightly polar, with tyrosine, which is neutral and polar, at codon 348 of the TGFBR1 protein (p.Cys348Tyr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Loeys-Dietz syndrome (PMID: 31475485). It has also been observed to segregate with disease in related individuals. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on TGFBR1 protein function.
