NM_000719.7(CACNA1C):c.5500G>A (p.Asp1834Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1834N variant (also known as c.5500G>A), located in coding exon 43 of the CACNA1C gene, results from a G to A substitution at nucleotide position 5500. The aspartic acid at codon 1834 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
