Uncertain significance for Familial hemiplegic migraine — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000702.4(ATP1A2):c.2335A>C (p.Ser779Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 779 of the ATP1A2 protein (p.Ser779Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with dystonia (PMID: 31737037). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ATP1A2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
