Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001190274.2(FBXO11):c.92C>A (p.Pro31Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 31 of the FBXO11 protein (p.Pro31Gln). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with FBXO11-related conditions. ClinVar contains an entry for this variant (Variation ID: 2971103). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Not Available". The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
