Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000722.4(CACNA2D1):c.1642G>C (p.Glu548Gln), citing Ambry Variant Classification Scheme 2023: The p.E548Q variant (also known as c.1642G>C), located in coding exon 19 of the CACNA2D1 gene, results from a G to C substitution at nucleotide position 1642. The glutamic acid at codon 548 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
