Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change affects DDX58 function (PMID: 28250012). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with DDX58-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.0009%). This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 96 of the DDX58 protein (p.Lys96Arg).
