Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001103.4(ACTN2):c.31A>C (p.Asn11His), citing Ambry Variant Classification Scheme 2023: The p.N11H variant (also known as c.31A>C), located in coding exon 1 of the ACTN2 gene, results from an A to C substitution at nucleotide position 31. The asparagine at codon 11 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
