Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003105.6(SORL1):c.2571+5_2571+11dup, citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs781363569, gnomAD 0.04%), and has an allele count higher than expected for a pathogenic variant. This sequence change falls in intron 18 of the SORL1 gene. It does not directly change the encoded amino acid sequence of the SORL1 protein. It affects a nucleotide within the consensus splice site. This variant has not been reported in the literature in individuals affected with SORL1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
