Uncertain significance for Carney-Stratakis syndrome; Paragangliomas with sensorineural hearing loss; Pheochromocytoma; Cowden syndrome 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003002.4(SDHD):c.479G>C (p.Ter160Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SDHD gene (transcript NM_003002.4) at coding-DNA position 479, where G is replaced by C. Submitter rationale: This sequence change disrupts the translational stop signal of the SDHD mRNA. It is expected to extend the length of the SDHD protein by 3 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SDHD-related conditions. This variant results in an extension of the SDHD protein. Other variant(s) that result in a similarly extended protein product (p.*160Lext*3) have been observed in individuals with SDHD-related disease (PMID: 24367056). This suggests that these extensions may be clinically significant. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
