NM_001723.7(DST):c.6775A>T (p.Arg2259Ter) was classified as Uncertain significance for Epidermolysis bullosa simplex 3, localized or generalized intermediate, with BP230 deficiency; Hereditary sensory and autonomic neuropathy type 6 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the C-terminus of the DST protein. Other variant(s) that disrupt this region (p.Leu2477Serfs*13, p.Tyr2366X) have been observed in individuals with DST-related conditions (PMID: 33471381). This suggests that this may be a clinically significant region of the protein. This variant has not been reported in the literature in individuals affected with DST-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Arg2259*) in the DST gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 391 amino acid(s) of the DST protein.
