NM_001283009.2(RTEL1):c.1262A>C (p.Tyr421Ser) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y421S variant (also known as c.1262A>C), located in coding exon 14 of the RTEL1 gene, results from an A to C substitution at nucleotide position 1262. The tyrosine at codon 421 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
