This variant lies in the CACNA1A gene (transcript NM_001127222.2) at coding-DNA position 286, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 96 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Glu96*) in the CACNA1A gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CACNA1A are known to be pathogenic (PMID: 10371528, 19486177, 25735478, 27250579). This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic.
