Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.5864A>G (p.Gln1955Arg), citing Ambry Variant Classification Scheme 2023. This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 5864, where A is replaced by G; at the protein level this means replaces glutamine at residue 1955 with arginine — a missense variant. Submitter rationale: The p.Q1955R variant (also known as c.5864A>G), located in coding exon 47 of the FBN1 gene, results from an A to G substitution at nucleotide position 5864. The glutamine at codon 1955 is replaced by arginine, an amino acid with highly similar properties. This variant was reported in individual(s) with thoracic aortic aneurysm/dissection (Arnaud P et al. Genet Med, 2019 Sep;21:2015-2024). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
