This variant lies in the RAG2 gene (transcript NM_000536.4) at coding-DNA position 361 through coding-DNA position 364, deleting 4 bases; at the protein level this means shifts the reading frame starting at threonine residue 121, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Thr121Phefs*9) in the RAG2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 407 amino acid(s) of the RAG2 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RAG2-related conditions. This variant disrupts a region of the RAG2 protein in which other variant(s) (p.Glu480*) have been determined to be pathogenic (PMID: 21624848, 29772310).
