Pathogenic for Distal myopathy with posterior leg and anterior hand involvement; Myofibrillar myopathy 5; Hypertrophic cardiomyopathy 26 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000007.14:g.128837160del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Gly201Valfs*51) in the FLNC gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FLNC are known to be pathogenic (PMID: 27908349). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with FLNC-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
