This variant lies in the TBC1D24 gene (transcript NM_001199107.2) at coding-DNA position 842, where C is replaced by A; at the protein level this means replaces serine at residue 281 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with tyrosine, which is neutral and polar, at codon 281 of the TBC1D24 protein (p.Ser281Tyr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive TBD1D24-related conditions (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TBC1D24 protein function with a negative predictive value of 80%.
