Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.1093A>G (p.Ile365Val), citing Ambry Variant Classification Scheme 2023: The p.I365V variant (also known as c.1093A>G), located in coding exon 7 of the FLNA gene, results from an A to G substitution at nucleotide position 1093. The isoleucine at codon 365 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on data from gnomAD, the G allele has an overall frequency of 0.0020% (4/202751) total alleles studied, with no hemizygote(s) observed. The highest observed frequency was 0.0219% (4/18238) of African/African American alleles. Based on the available evidence, the clinical significance of this variant remains unclear.
