Uncertain significance for Senior-Loken syndrome 8; Asphyxiating thoracic dystrophy 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_025132.4(WDR19):c.2562+19_2562+28del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WDR19 gene (transcript NM_025132.4) at 19 bases into the intron immediately after coding-DNA position 2562 through 28 bases into the intron immediately after coding-DNA position 2562, deleting this region. Submitter rationale: This sequence change falls in intron 22 of the WDR19 gene. It does not directly change the encoded amino acid sequence of the WDR19 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with WDR19-related conditions. ClinVar contains an entry for this variant (Variation ID: 2947058). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
