This variant lies in the GRIN2B gene (transcript NM_000834.5) at coding-DNA position 1223, where T is replaced by C; at the protein level this means replaces isoleucine at residue 408 with threonine — a missense variant. Submitter rationale: This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 408 of the GRIN2B protein (p.Ile408Thr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with GRIN2B-related conditions (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GRIN2B protein function.
