Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004990.4(MARS1):c.2323C>G (p.Pro775Ala), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MARS1 c.2323C>G (p.Pro775Ala) results in a non-conservative amino acid change located in the Methionyl-tRNA synthetase, anticodon-binding domain (IPR041872) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The frequency of this variant in the general population could not be determined as the technology used for large population databases (ExAC, gnomAD, ESP, 1000G) cannot detect variants of this type. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2323C>G in individuals affected with Severe early-onset pulmonary alveolar proteinosis due to MARS deficiency and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 2946133). Based on the evidence outlined above, the variant was classified as uncertain significance.
