Uncertain significance for GATA binding protein 1 related thrombocytopenia with dyserythropoiesis; Diamond-Blackfan anemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002049.4(GATA1):c.107C>T (p.Ser36Phe), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 36 of the GATA1 protein (p.Ser36Phe). This variant is present in population databases (rs782155672, gnomAD 0.001%). This variant has not been reported in the literature in individuals affected with GATA1-related conditions.
