Pathogenic for Bethlem myopathy 2; Ullrich congenital muscular dystrophy 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004370.6(COL12A1):c.8100+1G>C, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 52 of the COL12A1 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in COL12A1 cause disease. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individuals with autosomal dominant COL12A1-related myopathy (PMID: 29342313; Invitae). It has also been observed to segregate with disease in related individuals. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
