NM_030787.4(CFHR5):c.485_486dup (p.Glu163fs) was classified as Benign by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: CFHR5 c.485_486dupAA (p.Glu163LysfsX10) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, however current evidence is not sufficient to establish loss of function as a mechanism for disease. The variant allele was found at a frequency of 0.0068 in 281764 control chromosomes, predominantly at a frequency of 0.041 within the Non-Finnish European subpopulation in the gnomAD database, including 20 homozygotes. The observed variant frequency within Non-Finnish European control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in CFHR5. To our knowledge, no occurrence of c.485_486dupAA in individuals affected with CFHR5-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 294541). Based on the evidence outlined above, the variant was classified as benign.
