Pathogenic for Pheochromocytoma/paraganglioma syndrome 5; Mitochondrial complex II deficiency, nuclear type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004168.4(SDHA):c.351del (p.Asp117fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SDHA gene (transcript NM_004168.4) at coding-DNA position 351, deleting one base; at the protein level this means shifts the reading frame starting at aspartic acid residue 117, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with SDHA-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Asp117Glufs*11) in the SDHA gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SDHA are known to be pathogenic (PMID: 22974104, 24781757).
