Uncertain significance for Hereditary spastic paraplegia 57; Hereditary motor and sensory neuropathy, Okinawa type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006070.6(TFG):c.1017T>A (p.Tyr339Ter), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with TFG-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr339*) in the TFG gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 62 amino acid(s) of the TFG protein.
