This variant lies in the CHCHD10 gene (transcript NM_213720.3) at coding-DNA position 364, where T is replaced by C; at the protein level this means replaces cysteine at residue 122 with arginine — a missense variant. Submitter rationale: This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 122 of the CHCHD10 protein (p.Cys122Arg). This variant is present in population databases (rs763453989, gnomAD 0.003%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change affects CHCHD10 function (PMID: 29789341).
