NM_000834.5(GRIN2B):c.2352_2359+2dup was classified as Pathogenic for Developmental and epileptic encephalopathy, 27; Intellectual disability, autosomal dominant 6 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the GRIN2B gene (transcript NM_000834.5) at coding-DNA position 2352 through the canonical splice donor site of the intron immediately after coding-DNA position 2359, duplicating this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has been observed in individual(s) with clinical features of GRIN2B-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change falls in intron 11 of the GRIN2B gene. It does not directly change the encoded amino acid sequence of the GRIN2B protein. It affects a nucleotide within the consensus splice site.
