This variant lies in the VCP gene (transcript NM_007126.5) at coding-DNA position 467, where G is replaced by T; at the protein level this means replaces glycine at residue 156 with valine — a missense variant. Submitter rationale: This variant disrupts the p.Gly156 amino acid residue in VCP. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 23868359, 32036797; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
