Uncertain significance for Lower motor neuron syndrome with late-adult onset; Frontotemporal dementia and/or amyotrophic lateral sclerosis 2; Autosomal dominant mitochondrial myopathy with exercise intolerance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_213720.3(CHCHD10):c.410-3C>G, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CHCHD10 gene (transcript NM_213720.3) at 3 bases into the intron immediately before coding-DNA position 410, where C is replaced by G. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has not been reported in the literature in individuals affected with CHCHD10-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change falls in intron 3 of the CHCHD10 gene. It does not directly change the encoded amino acid sequence of the CHCHD10 protein. It affects a nucleotide within the consensus splice site.
