NM_139276.3(STAT3):c.2137G>T (p.Val713Leu) was classified as Pathogenic for STAT3 gain of function; Hyper-IgE recurrent infection syndrome 1, autosomal dominant by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 713 of the STAT3 protein (p.Val713Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of hyper-IgE syndrome (PMID: 18602572, 25962528; Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt STAT3 protein function with a positive predictive value of 80%. This variant disrupts the p.Val713 amino acid residue in STAT3. Other variant(s) that disrupt this residue have been observed in individuals with STAT3-related conditions (PMID: 22751495, 25739182), which suggests that this may be a clinically significant amino acid residue. For these reasons, this variant has been classified as Pathogenic.
