Pathogenic for Familial temporal lobe epilepsy 7; Norman-Roberts syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005045.4(RELN):c.6526C>T (p.Gln2176Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RELN gene (transcript NM_005045.4) at coding-DNA position 6526, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 2176 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with RELN-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln2176*) in the RELN gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RELN are known to be pathogenic (PMID: 10973257, 26046367, 28454995).
