Uncertain significance for Marfan syndrome; Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000138.5(FBN1):c.5129T>C (p.Leu1710Ser), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt FBN1 protein function. This sequence change replaces leucine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1710 of the FBN1 protein (p.Leu1710Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with acromicric dysplasia (PMID: 33082559). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
