Submitter rationale: The c.440G>A variant in MYOC is a missense variant predicted to cause substitution of Arginine by Glutamine at amino acid 147 (p.Arg147Gln). The highest minor allele frequency of this variant was in the Ashkenazi Jewish genetic ancestry group of gnomAD (v4.1.0) = 0.008748, which met the ≥ 0.001 threshold set for BS1 (259 alleles out of 29,606, meeting the threshold of ≥ 5 of at least 2,000 observed alleles). The REVEL score = 0.082, which was within the 0.017-0.183 range for BP4_Moderate, suggesting that the variant does not impact MYOC function.
