NM_004415.4(DSP):c.3346_3354del (p.Tyr1116_Ile1118del) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 3346 through coding-DNA position 3354, deleting 9 bases. Submitter rationale: The c.3346_3354delTATGAGATT variant (also known as p.Y1116_I1118del) is located in coding exon 23 of the DSP gene. This variant results from an in-frame TATGAGATT deletion at nucleotide positions 3346 to 3354. This results in the in-frame deletion of three amino acids (YEI) a at codons 1116 to 1118. This variant has been observed in at least one individual with a personal and/or family history that is consistent with dilated cardiomyopathy (Ambry internal data). These amino acid positions are conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
