Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_024426.6(WT1):c.578C>T (p.Ala193Val), citing Ambry Variant Classification Scheme 2023: The p.A188V variant (also known as c.563C>T), located in coding exon 1 of the WT1 gene, results from a C to T substitution at nucleotide position 563. The alanine at codon 188 is replaced by valine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
