This variant lies in the WT1 gene (transcript NM_024426.6) at coding-DNA position 578, where C is replaced by T; at the protein level this means replaces alanine at residue 193 with valine — a missense variant. Submitter rationale: This missense variant replaces alanine with valine at codon 188 in the WT1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with a horsehoe kidney, dysmorphic features, and a ventricular septal defect (PMID: 31738409). This variant has been identified in 2/244504 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
