This variant lies in the MARS1 gene (transcript NM_004990.4) at coding-DNA position 1531 through 14 bases into the intron immediately after coding-DNA position 1539, deleting this region. Submitter rationale: This variant results in the deletion of part of exon 12 (c.1531_1539+14del) of the MARS gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in MARS cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MARS-related conditions.
