Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.6337T>C (p.Tyr2113His), citing Ambry Variant Classification Scheme 2023: The p.Y2113H variant (also known as c.6337T>C), located in coding exon 51 of the FBN1 gene, results from a T to C substitution at nucleotide position 6337. The tyrosine at codon 2113 is replaced by histidine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
