Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_016169.4(SUFU):c.910G>A (p.Asp304Asn), citing Ambry Variant Classification Scheme 2023: The c.910G>A variant (also known as p.D304N), located in coding exon 7 of the SUFU gene, results from a G to A substitution at nucleotide position 910. The amino acid change results in aspartic acid to asparagine at codon 304, an amino acid with highly similar properties. However, this change occurs in the last base pair of coding exon 7, which makes it likely to have some effect on normal mRNA splicing. This nucleotide position is highly conserved in available vertebrate species. This amino acid position is highly conserved in available vertebrate species. In silico splice site analysis for this alteration is inconclusive. However, RNA studies have demonstrated that this alteration does not result in abnormal splicing in the set of samples tested (Ambry internal data).
