Uncertain Significance for Von Hippel-Lindau syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000551.4(VHL):c.-39_-10del, citing ACMG Guidelines, 2015. This variant lies in the VHL gene (transcript NM_000551.4) at 39 bases upstream of the translation start (5' untranslated region) through 10 bases upstream of the translation start (5' untranslated region), deleting this region. Submitter rationale: This variant causes a 30-nucleotide deletion in the 5' untranslated region of the VHL gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with VHL-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
