Uncertain significance for Autosomal dominant nonsyndromic hearing loss 3B; Hidrotic ectodermal dysplasia syndrome; Autosomal recessive nonsyndromic hearing loss 1B; Autosomal recessive nonsyndromic hearing loss 1A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001110219.3(GJB6):c.460T>A (p.Phe154Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 154 of the GJB6 protein (p.Phe154Ile). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with GJB6-related conditions (PMID: 29739340). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt GJB6 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
