This variant lies in the ANO5 gene (transcript NM_213599.3) at coding-DNA position 1013, where G is replaced by A; at the protein level this means replaces serine at residue 338 with asparagine — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 338 of the ANO5 protein (p.Ser338Asn). This variant also falls at the last nucleotide of exon 10, which is part of the consensus splice site for this exon. This variant is present in population databases (rs766507413, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with ANO5-related conditions. ClinVar contains an entry for this variant (Variation ID: 2926268). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
