Pathogenic for Marfan syndrome; Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000138.5(FBN1):c.3464-6C>A, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 28 of the FBN1 gene. It does not directly change the encoded amino acid sequence of the FBN1 protein. For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has been observed in individual(s) with clinical features of Marfan syndrome (PMID: 24161884; Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency).
