This variant lies in the NPR2 gene (transcript NM_003995.4) at coding-DNA position 844, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 282 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Gln282*) in the NPR2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NPR2 are known to be pathogenic (PMID: 15146390, 15572448, 16384845). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with acromesomelic dysplasia (PMID: 15146390).
